Just Ban the User Who is Spamming...
You can add random questions peple have to answer everytime they are posting a new topic.
ENABLE Captcha.
Image Verification.
Email Confirmation.
IP Blocking is worth a try.
Swear filters.

Here's what i do and it helps me with 95% spam.
1) use the htaccess as Sankvid mentioned.
2) for vB use Advanced forum rules to trap bots who don't accept the rules.
3) Use spam-o-matic. at first i was skeptical about this, but after alot of monitoring this addon it blocked 90% bot registrations. and best of all, if someone gets through it will put their posts in moderation queue.
Now, Do note that some times people register and then use a multiposter.
they will get through and if you have spam-o-matic it will nab their ass.

Since I opened Taboo Girls I only had problems with two spammers. And solved them quickly, lol. Basically, I'm using Q&A verification system, and newly registered users can't post anywhere except in the General Discussion Area, where after they make 2 posts, they're automatically promoted to the regular member usergroup. Which allows them posting of pics n videos.
I've blocked bunch of temporary email services, Q&A verification is enabled for the probation users, and they have to enter the answer to submit the post/thread.
It works like a charm...

I agree. I avoid most forums that use autoposters. it looks so messy and awful! it's like 100 threads on the latest S03E05 All on FS or Filesonic. It's useless. it'd be good if all were on different hosts or something with variety but it's practically the same thing times 100 every single day something new comes out!

i have seen many threads in community cooperative section where forum owners are requesting users to add their forum to multi poster
well now the main forum whose url i include in links,i upload many files n post them so am i spamming there?
i dont post in mega thread there as he submit my links so for that i cant make mega thread
i have mega threads on exbii,porn-w,intporn n final4ever where i post manually,jayfella's multi poster has a bug in replying to existing content thats y cant make a mega thread using MP5 on all forums
You may very well have but please re-read my reply. I said you'd never find a WEBMASTER requesting this not a forum owner. The line above that using != is a coders symbol meaning not equal to.
Webmasters have a certain level of intelligence. Anyone can buy a forum becoming a forum owner but it doesn't automatically make them a webmaster implying: forum owner != webmaster.
jasb007 is a WEBMASTER and not just a Forum Owner which is why he's requesting this.
Anyway we still don't know what forum script he's using?
That's pretty important as their are mods available and things you can do for different scripts.
Ip bans + enable captcha and that is it, even if there are some that still get through you will still see the difference in posts and purge them out or just ban the member and leave the posts. 
But i would i advise you to check carefully which posts you delete and which multiposter members you ban because some of them post really quality stuff and lately most of the posters do it threw the multiposter instead of manual posting and you might lose a lot of good members that way.
Try not to ban the multiposters that are posting in sections like music/movies/mobiles/tv-shows because not a lot of members post there and those sections are really important because we visit them frequently.
