You can create that kind of sites by using scripts developed by yourself or You can Buy any filehosting scripts.
xFilesharing is the most popular file hosting script.
You also need a good Dedicated server with lots of Space. Because without lots of space you cant create a hosting site.
And you need lots of money to get those basic stuff to create file hosting sites.
And you must prepare to face some losses at some of first months.

You need:
1. Money (for the initial investment) 'cause you need to pay...
a. Personnel - To to do everything, 'cause you don't know the slightest thing (first, to create all, and then, to maintain it {and it is not cheap}). That of course includes the one in-charge who answers the phone or email of angry users.
b. Equipment - servers with all the hardware they need (hard disks, routers, cables, coolers... etc). And everything an office needs.
c. A place - Expenses include electric, phone and internet bills. (It can't be your garage)
d. Software - It can be free or commercial, you decide.
e. An extremely good internet connection - 'cause you're supposed to download and upload lots of people at the same time. (can't do it with your home basic internet)
2. You have to work competing with others filehostings, so you need some really good marketing to beat some of them. Maybe paying the uploaders or giving some benefits for the free accounts. (yes, you're not getting ANY MONEY at the beginning, only debts and more debts)
3. A good lawyer in case someone upload something absolutely and completely illegal that makes FBI search you with satellites and break down your door in the middle of the night.
Also he/she must be someone you can trust, 'cause all the great hollywood franchises (and others too) love to demand for millions (or billions) and they don't hesitate to pay a little something to your lawyer (5 or 6 zeros), just to win easy.
